We present here a new biodrug dedicated to CRC prevention among high risk population. This biodrug is composed of a food-grade bacterium (Propionibacterium freudenreichii - Pf) as a vector, and the TRAIL (TNF related apoptosis inducing-ligand) protein.

Vector : Pf (wild-type) can be used as a tool for addressing or targeting substances to the intestine. The advantage is their survival time up to two weeks compared to only two or three days for lactic acid bacteria Moreover, Pf triggers the intrinsic death pathway throught the production of SCFA (short chain fatty acid), therefore showing an effect on inhibiting the formation or growth of tumors. This can be achieved by a pro-apoptotic, anti-proliferative, cytotostatic or cytotoxic effect.

Protein of interest: The TRAIL protein is known to induce the death of many tumor cells by triggering the extrinsic death pathway In addition, it has been proven that it also inhibits the growth of xenografted colon tumors in nude mice.

Synergy: Therefore, Pf bacterium expressing TRAIL protein triggers two different pathways, meanwhile creating a synergistic effect on tumor cells
