Innovative strategy to improve stability and permeability of therapeutic peptides

Context
Peptides play a critical role in various biological functions, but their use in therapeutic applications faces many challenges. Peptides are actually degraded very rapidly in the organism by peptidase, which prevents them from reaching their biological target. In addition, the cell membrane constitutes an impenetrable barrier against most macromolecules, including peptides. The current stakes for the development of therapeutic peptides are based both on improving their ability to cross through cell membranes, and on increasing their metabolic stability by protecting them against enzymatic breakdown.

Benefits
This project has developed an innovative strategy that enables both increasing the resistance of a peptide to enzymatic breakdown, while also improving its membrane permeability. In addition, this strategy does not disturb the peptide’s secondary structure, which thus preserves its biological activity. This strategy applies to peptides as well as all other compounds bearing an amine function.
