DESCRIPTION
Nanoparticles are immobilized on a specific material for the supported synthesis of biomolecules such as nucleic acid fragments. Thanks to this material, a good functionalisation of the nanoparticles is obtained and synthesized biomolecules are of good quality. Each nanoparticle can be functionalised with two different biomolecules and/or two different labels, allowing for example mix of fluorescent and radioactive detection.

STAGE OF DEVELOPMENT
The laboratory masters nanoparticles grafting on the specific material, synthesis of oligonucleotides, incorporation of functionalities and release after functionalisation.
A recognition probe of hepatitis B virus genome and a RNA fragment targeting the MMP9 tumour marker protein were synthesized. Quality was validated by HPLC and in vitro recognition specificity by ELOSA (enzyme-like oligo-sorbent assay) and SPR (surface plasmon resonance)

INTELLECTUAL PROPERTY
Patent pending in FR
Priority date: July 12, 2011 (FR11 56338)

ADVANTAGES / NOVELTY
• Higher sensitivity and selectivity thanks to the high number of grafted biomolecules by surface area unit (0,1-2 oligonucleotides/nm²).
• Versatile strategy for the synthesis of a large panel of biomolecules and functionalities.
• Orientation’s control of grafted biomolecules.
• Excellent stability of conjugated nanoparticles in solution (PBS buffer or water) for several weeks.
• Good grafting’s rate of biomolecules.
• Good quality of synthesized biomolecules (DNA, RNA).
• High in vivo stability

APPLICATIONS
Grafting of nanoparticles according to demand specifications for:
• Imaging (contrast agent)
• Diagnosis
• Therapy
• Vaccines
